瘦素及瘦素受体在胃癌组织和胃癌细胞株中的表达

目的]探讨胃癌细胞株和胃癌组织是否有瘦素和瘦素受体表达,以及瘦素对胃癌细胞株的增殖作用.方法]免疫组化和RT-PCR检测胃癌细胞株及胃癌组织中瘦素和瘦素受体表达,MTT法测定瘦素对胃癌细胞株SGC7901、MGC803、MKN45的增殖作用.结果]胃癌细胞株SGC7901、MGC803及MKN45均有瘦素及瘦素受体表达;47例胃癌组织石蜡切片标本检测有瘦素表达20例,有瘦素受体表达23例,两者共同表达17例;19例新鲜胃癌组织标本,有瘦素mRNA表达13例,瘦素受体mRNA表达12例,两者共同表达10例;与对照组相比,100ng/mL的瘦素可使胃癌细胞株SGC7901、MGC803、MKN45的增殖率分别为1.45、1.56及1.54.结论]胃癌细胞株及胃癌组织普遍存在瘦素和瘦素受体表达,瘦素可促进胃癌细胞株增殖.

Expression of Leptin and Leptin Receptor in Gastric Cancer Cell Line and Gastric Cancer Tissue

Objective To investigate the expression of the leptin and leptin receptor in gastric can-cer cell line and gastric cancer tissue and the proliferative effect of leptin in gastric cancer cell line. Methods The expression of leptin and leptin receptor was detected with the immunohistochemistry and RT-PCR respectively. The proliferative effect of leptin was investigated with MTT assay. Results Leptin and leptin receptor were expressed in gastric cancer cell line and gastric cancer tis-sue. Among 47 paraffin section specimen 20 specimen expressed leptin , 23 specimen expressed lep-tin receptor and 17 specimen expressed both of them through immunohistochemical staining. Among 19 fresh gastric cancer specimen, 13 specimen expressed leptin mRNA, 12 specimen expressed leptin receptor mRNA and 10 specimen expressed both of them through RT-PCR. Compared with control group, the proliferation rate of gastric cancer cell line SGC7901,MCG803 and MKN45 with 100 ng/mL leptin in cell culture medium was 1.45,1.56 and 1.54 respectively. Conclusions Leptin and leptin receptor are expressed in gastric cancer cell line and gastric cancer tissue. Leptin can pro-mote the proliferation of gastric cancer cell line.
