不同日龄新生儿茶碱药动学研究

 目的 研究不同日龄新生儿茶碱药动学。方法 采用荧光偏振免疫分析法测定20例新生儿茶碱血药浓度，氨茶碱剂量按5 mg·kg^-1恒速静滴。经时采样，测得数据，用3P87程序处理数据，以一室开放模型拟合，A组为7 d以内早产儿，B组为7 d以内的足月儿，C组为8例8～27 d足月儿。结果 药动学参数Ke,t_1/2，Vd和CL分别为：A组(0.031±0.007)h^-1，(23±5) h,(0.82±0.22)L·kg^-1，(25±8)ml·h^-1·kg^-1；B组(0.029±0.006)h^-1，(25±5)h，(0.86±0.13)L·kg^-1，(25±6)ml·h^-1·kg^-1；C组(0.040±0.008)h^-1，(18±3)h,(0.78±0.14)L·kg^-1,(31±5)ml·h^-1·kg^-1。结论 A组与C组，B组与C组的Ke,t_1/2值经方差分析有显著性差异(P＜0.05)。 

Study on pharmacokinetics of theophylline in different groups of infants

OBJECTIVE To study the pharmacokinetics of theophylline in different groups of infants. METHODS The serum concentrations of theophylline were determined in 20 cases of infants by fluorescence polarization immunoassay.Group A was 1～7 day old premature;Group B was 1～7 day old term neonate;Group C was 8～27 day old term neonate. RESULTS The values of Ke,t_1/2,V_dand Cl were (0.031±0.007) h^-1，(23±5) h,(0.82±0.22) L·kg^-1，(25±8) ml·h^-1·kg^-1,respectively in group A;(0.029±0.006)h^-1，(25±5) h，(0.86±0.13)L·kg^-1，(25±6) ml·h^-1·kg^-1in group B;(0.040±0.008) h^-1，(18±3) h,(0.78±0.14) L·kg^-1,(31±5) ml·h^-1·kg^-1in group C. CONCLUSION The values of Ke,t_1/2of group A and group B were significantly different from group C (P＜0.05).
