摘    要:目的 : 观察二十二碳六烯酸 ( DHA)对神经细胞生长发育的作用。方法 : 将无血清培养的新生大鼠大脑神经细胞分为实验组和对照组 ,实验组加入 0 .33mol/ L( 5mol/ 1 5L)乳化DHA40 μl。对两组细胞进行神经细胞活力、蛋白质含量、神经特异性烯醇化酶 ( NSE)活性测定及神经细胞形态学观察。结果 : 实验组大脑神经细胞突起生长速度、胞体面积和直径、神经细胞存活率、NSE和神经细胞活力及蛋白含量均显著高于对照组。结论 : DHA对神经细胞的生长有促进作用 ,且这一作用可能与促进神经细胞蛋白质合成有关。

Abstract:Objective:To study the effects of DHA on the growth and development of cerebral neurons. Methods:The primary culture of the cerebral neurons of newborn rats was carried out in vitro by using serum free medium. Cells were divided into two groups, experimental and control. DHA was added to the medium in the experimental group. After the neurons were cultured, the morphological states of the neurons were investigated; the positive rate of MTT、 NSE activity and protein content were measured. Results:Both the number of survival cells and the outgrowth of neurites were increased by DHA. In the presence of DHA, the diameter and volume of neurons as well as the content of protein and the activity of NSE and MTT positive rate were markedly increased in image analysis. Conclusion:It is suggested that DHA may play a promotive role in the growth and development of cerebral neurons.
