泌乳素对人B淋巴细胞HLA-DR表达的影响

摘    要:目的探讨泌乳素(PRL)与B淋巴细胞功能的关系。方法以生理浓度(10 ng.ml-1)的人PRL刺激培养IM-9细胞系,并设置未刺激组、脂多糖(LPS)刺激组、PRL刺激组、LPS+PRL刺激组、LPS+溴隐停(Brc)刺激组和LPS+PRL+Brc刺激组;采用免疫细胞化学S-P染色法检测不同刺激组B淋巴细胞HLA-DR的表达情况。结果LPS刺激组和PRL刺激组HLA-DR的平均光密度(MOD)与未刺激组相比有明显增加(P<0.05);LPS+PRL刺激组HLA-DR的MOD与LPS、PRL刺激组相比明显增加(P<0.01);LPS+Brc刺激组HLA-DR的MOD与LPS刺激组与LPS+PRL+Brc刺激组相比均明显降低(P<0.05)。结论PRL对B淋巴细胞有活化作用。

Abstract:Objective To study the relationship between prolactin and B lymphocyte.Methods Cultivated IM-9 cells were stimulated by human prolactin(10 ng·ml~(-1)),and were divided into non-stimulated group,LPS-stimulated group,PRL-stimulated group,LPS & PRL-costimulated group,LPS & Brc(bromocrytine)-costimulated group and LPS & PRL & Brc-costimulated group.The expression of HLA-DR of each group was detected by streptavdin-peroxidase-biotin(S-P) of immunocytochemical methods.Results The mean optic density(MOD) of LPS-stimulated and PRL-stimulated groups were obviously higher than that of non-sitimulated group (P<0.05).The MOD of LPS & PRL-costimulated group was obviously higher than that of LPS-stimulated group and PRL-stimulated group(P<0.01).The MOD of LPS & BRC-cositimulated group was significantly lower than that of the LPS-sitimulated group and LPS & PRL & BRC group(P<0.05).Conclusion PRL plays an important role in the activation of B lymphocytes.
