肺癌组织p300/CBP基因突变机制的研究

目的探讨p300/CBP基因在肺癌中的发生、发展中的作用.方法采用PCR-SSCP及DNA序列分析技术检测30例新鲜肺癌组织p300/CBP的基因突变,以30例正常肺组织作对照.结果肺癌组织中p300/CBP基因存在1个位点的点突变,位于p300gDNA的4773碱基,T→G,突变点为编码区,丝氨酸(Ser)→精氨酸(Arg).本组30例肺癌组织中,有1例突变,结论肺癌组织中p300/CBP基因存在1个位点的点突变.

Study the mutation of p300/CBP Gene in lung cancer

Objective To study the role of mutation of p300/CBP gene in lung cancer. Methods Mutation analysis of the p300/CBP gene were carried out in 30 cases of fresh lung cancer samples using a combination of polymerase chai n reaction single-strand conformation polymorphism (PCR-SSCP) and DNA sequencing , contrasted with their corresponding normal tissues. Results p300/CBP genes mutations were detected in 1 of 30 fresh LCs.It is point mutations, T to G in n ucleotide 4773 respectively in genomic DNA of P300. It is coding area. It leads to the change of the amino acids, which is Ser to Arg. Of 30 patients, we discov ed 1 cases with the single site mutation. Conclusion One Point mutation of p 300/CBP gene is presented in lung cancer.
