人脑胶质瘤中MMP-2与MMP-9酶活性改变和蛋白的表达及其意义

摘    要:目的:探讨基质金属蛋白酶2(matrix metalloproteinase 2,MMP-2)与基质金属蛋白酶9(matrix metallopro-teinase 9,MMP-9)的表达与人脑胶质瘤恶性行为之间的关系。方法:采用明胶酶谱与免疫组化方法对50例人脑胶质瘤和4例正常脑组织中的MMP-2及MMP-9进行检测。结果:各级别组中,MMP-2和MMP-9的酶原形式无差别;在正常、Ⅰ级和Ⅱ级的胶质瘤组织中无活性酶存在,在Ⅲ级和Ⅳ级胶质瘤组织中,MMP-2活性酶的含量分别为42.586±10.245A.mm2/g.L和62.983±6.454A.mm2/g.L,MMP-9酶的含量分别为74.559±8.398A.mm2/g.L和176.117±11.922A.mm2/g.L,经免疫组化染色,从正常脑组织到Ⅳ级胶质瘤组织,MMP-2的阳性率分别为25%,0,23.8%,70%,87.5%;MMP-9的阳性率为0,33.3%,19%,60%,87.5%,MMP-2,9的酶含量及阳性率在各级别中的差异具有统计学意义。结论:在人脑胶质瘤中,MMP-2和MMP-9酶的活性变化与其病理分级密切相关,可作为判断胶质瘤恶性程度的重要参考指标。

Abstract:Objective: To investigate the relationship between the expressions and enzymatic activities of type IV collagenase(including matrix metalloproteinase 2,MMP-2 and matrix metalloproteinase 9,MMP-9) and the malligant biogical behavior of human brain gliomas.Methods: The enzymatic activities of MMP-2 and MMP-9 were detected by gelatin zymography in 50 cases of human brain gliomas and 4 cases of normal human brian tissues.Immunohistochemical study was used for MMP2 and MMP-9 expression in all cases.Results: In gelatin zymagraphy,the latent zymogen forms of MMP-2 and MMP-9 were not different among the four grades of gliomas.In normal brain tissues and grade Ⅰ,Ⅱ gliomas tissue,there was no zymogen form of MMP-2 and MMP-9.In grade III and Ⅳ gliomas,the counts of the enzymatic activities of MMP-2 were separately 42.586±10.245 A.mm~2/g.L and 62.983±6.454 A.mm~2/g.L;and the counts of the enzymatic activities of MMP-9 were separately 74.559±8.39 A.mm~2/g.L and 176.117±11.922 A.mm~2/g.L.After Immunohistochemical study,from normal human brian tissues to grade Ⅳ glioma tissues,the positive stained-cell rates of MMP-2 were separately 25%,0,23.8%,70% and 87.5%;the positive stained-cell rates of MMP-9 were separately 0,33.3%,19%,60% and 87.5%.The counts of the enzymatic activities and positive stained-cell rate of MMP-2 and MMP-9 between the low-grade and high-grade tumors were statistically significant.Conclusion: In human brain gliomas,the change of MMP-2 and MMP-9 is associated with the malignant progression;matrix metalloproteinase 2;matrix metalloproteinase 9which were useful factors to judge the malignant degree of gliomas.
