酒精性肝损伤大鼠肠道屏障功能改变及谷氨酰胺的保护作用观察

摘    要:目的 观察酒精性肝损伤大鼠肠道屏障功能的改变,并探讨谷氨酰胺的保护作用。方法 36只SD大鼠随机分为正常对照组(A组)、模型组(B组)和谷氨酰胺治疗组(C组),分别检测肠道细菌易位率、肠粘膜通透性,同时观察血生化、肝脏和末段回肠粘膜病理学改变。结果 与A组比,B组大鼠血清AST和ALT水平显著升高(AST:256.6±50.8U/L对175.2±16.2U/L,P〈0.05;ALT:86.5±5.4U/L对53.7±13.2U/L,P〈0.05),C组无显著变化(P〉0.05);B组肠道细菌易位率明显增高(62.5%,P〈0.05),C组无显著变化(35.0%,P〉0.05);B组肠粘膜通透性显著升高(235.1±26.4ml·min^-1·cm^2对30.1±33.6ml·min^-1·cm^2,P〈0.05),C组肠粘膜通透性比B组明显下降(190.9±19.8ml·min^-1·cm^2对235.1±26.4ml·min^-1·cm^2,P〈0.05):B组大鼠末段回肠病理损伤明显加重(3.8±0.9对0.4±0.5,P〈0.05),C组比B组明显下降(2.7±0.7对3.8±0.9,P〈0.05)。结论 大鼠酒精性肝损伤伴有肠道屏障功能改变,谷氨酰胺对大鼠酒精性肝损伤及肠道屏障功能具有双重保护作用。

Protection of glutamine on intestinal barrier function in ethanol-induced liver injury in rats

Abstract:Objective To investigate the changes of intestinal barrier function and protection of glutamine on chronic ethanol-induced liver injury in rats. Methods 36 SD rats were divided randomly into group A (normal control), group B (model) and group C (glutamine-treated) and the changes in liver biochemistry, intestinal mucosal permeability,bacterial translocation and intestinal mucosa pathohistology were detected,respectively. Results The serum AST(256.6±50.8 U/L vs. 175.2±16.2 U/L,P〈0.05) and ALT levels(86.5±5.4 U/L vs. 53.7±13.2U/L,P〈0.05),the intestinal mucosal permeability (62.5% vs. 0.0%,P〈0.05),and the bacterial translocation rate of intestinal tract (235.1±26.4 ml·min^-1·cm^2 vs. 30.1±33.6 ml·min^-1·cm^2,P〈0.05) increased obviously,and the pathological injury of intestinal mucosa (3.8±0.9 vs 0.4±0.5, P〈0.05) aggravated in group B as compared to in group A;while all the parameters in group C were alleviated as compared to in group B. Conclusion In experimental ethanol-induced liver injury in rats,glutamine has a protective role in liver and intestinal barrier injuries.
