复方当归注射液对肌成纤维细胞增殖和凋亡影响的研究

摘    要:目的探讨复方当归注射液对肌成纤维细胞增殖和凋亡的影响,为其治疗口腔黏膜下纤维性变提供理论依据。方法体外培养人口腔黏膜成纤维细胞,并使其转分化为肌成纤维细胞,采用MTT法检测复方当归注射液对成纤维细胞及肌成纤维细胞的增殖抑制作用,并比较两者的抑制率有无差别。采用流式细胞术检测不同浓度的复方当归注射液作用于肌成纤维细胞后不同时间的凋亡率。结果复方当归注射液对成纤维细胞与肌成纤维细胞均有抑制作用,存在时间和浓度依赖性,对肌成纤维细胞的增值抑制作用要强于成纤维细胞,体现了该药物对肌成纤维细胞具有一定的靶向作用。540 mg·mL-1的复方当归注射液作用于肌成纤维细胞细胞24、48、72 h,其凋亡率可随时间、浓度的增加而增加,具有时间和浓度的依赖性。各浓度组与对照组相比及各浓度组之间相比均有统计学意义(P<0.01)。结论复方当归注射液可靶向性抑制肌成纤维细胞增殖,从而具有治疗口腔黏膜下纤维性变的可能性。

Abstract:Objective To determine the effect of compound angelica injection on the proliferation and apoptosis of myofibroblasts, and explore the mechanism of these agents to effectively treat oral submucous fibrosis. Methods The effect of compound angelica injection on fibroblasts and myofibroblasts were detected by MTT assay. Cell apoptosis percentage at different time and concentration of compound angelica injection were determined by fl ow cytometsy. Results Compound angelica injection inhibited the proliferation of fi broblasts and myofi broblasts in a dose and time dependent manner, and the inhibition rate of myofi broblasts was higher than that of fi broblasts. Compound angelica injection(5- 40 mg·mL- 1) was administered individually for 24, 48 and 72 h, and the apoptosis rate of myofi broblasts slightly increased with time and concentration with statistical signifi cance(P 〈0.01). These agents markedly improved the apoptosis rate in a time and concentration dependent manner. Conclusion Compound angelica injection can inhibit proliferation of myofi broblasts as a target. It may be a candidate for the treatment of oral submucous fi brosis.
