| 摘 要： | 目的探讨PC12细胞缺糖损伤时应激基因葡萄糖调节蛋白(grp75)的表达变化.方法无葡萄糖培养基培养PC12细胞建立细胞缺糖损伤细胞模型,用RT-PCR、Western blot 和细胞免疫化学法分析缺糖损伤时grp75的表达情况.结果 RT-PCR结果显示细胞缺糖损伤3h起grp75基因转录水平已显著增高,并在缺糖后3～24h间持续上升;蛋白印迹法和细胞免疫化学法证实grp75在细胞缺糖过程中蛋白水平的表达明显高于对照组,且随着缺糖时间推延而进一步升高.结论缺糖损伤时,细胞中的应激基因grp75特异性上调表达.

| Abstract: | Objective: To explore the expression of grp75 in PC12 cell under glucose deprivation. Methods: PC12 cells were cultured with glucose free medium, and the expression of grp75 was determained by RT-PCR,Western blot and immunocytochemistry. Results: The expression of grp75 was upreglated since PC12 cells was glucose deprived for 3h,and the expression was increased markedly during the process of glucose derivation. Conclusion: grp75 was upreglated to generate cytoprotective effect under glucose deprivation. |
