| 摘 要： | 目的：探讨大鼠嗅球成鞘细胞（Olfactory cnsheating cells,OECs）分离与培养的方法。方法：无菌条件下嗅球置于少量人工脑脊液中，用眼科镊子直接将嗅球撕烂、挤压，得片状组织块，胰酶消化，吹打，细胞悬液进行培养。结果：本方法分离、培养的OECs具有双极或多极突起，且突起十分细长，神经生长因子受体和胶质纤维酸性蛋白免疫组化呈阳性。结论：该方法简便易行，为深入研究OECs神经生物学特性、阐明促进中枢神经元轴突生长的作用机制打下基础。

| Abstract: | Objective To investigate an effective method of isolation, culture, characterzation of ensheathing cells from rat olfactory bulbs. Methods Olfactory bulbs of neonatal rats were put into ACSF, and torn and extruded into pieces by forceps. After being digested with pancreatin, the cell suspension was gathered and cultured. Results Cultured olfactory ensheating cells (OECs) appeared with bipolar or multipolar slender processes. p75NGFR and GFAP immunocytochemistry was positive. Conclusion The reported method is simple and easy and is foundamental for further research on the biological characteristics of OECs and the mechanism of enhancement of neurona growth in CNS. |
