羧甲基葡聚糖体外抗肿瘤作用的研究

摘    要:目的研究不同取代度羧甲基葡聚糖(CMG)的体外抗肿瘤作用。方法以HL-7702正常肝细胞、HepG-2肝癌细胞、MGC-803胃癌细胞为实验材料,通过四甲基偶氮唑盐(MTT)比色法分析。结果各种取代度的CMG对HL-7702的生长影响较小,但对HepG-2和MGC-803的抑制作用明显,并且随浓度升高对HepG-2、MGC-803的抑制作用也随之增强,尤以取代度为0.556的CMG2最为显著。CMG2浓度达到500μg/mL时对HepG-2的生长抑制率为54.3%,浓度为1 000μg/mL时对MGC-803抑制率达到51.3%。结论在体外实验中,取代度0.55左右的CMG能有效抑制HepG-2肝癌细胞、MGC-803胃癌细胞的生长。

Abstract:Purpose To detect the anti-cancer effect of carboxymethylg-lucan(CMG) with different degree of substitution(DS)in vitro.MethodsUsing HL-7702,HepG-2 and MGC-803 cell as the experimental objects,the anti-cancer effect of CMG was studied by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT) colorimetric Method.ResultsCMG weakly inhibited the growth of HL-7702,but obviously affected the growth of HepG-2 and MGC-803.The higher concentration of CMG,the bigger inhibitory effect on HepG-2 and MGC-803 cell is.The inhibitory ratio of CMG2 to HepG-2 was 54.3% at the concentration of 500 μg/mL,to MGC-803 was 51.3% at the concentration of 1 000 μg/mL.ConclusionCMG(DS≈0.55) could effectively inhibit the growth of HepG-2 and MGC-803 cell.
