| 摘 要： | 目的观察APE1siRNA在KM3细胞内表达情况.方法将构建的MM特异APE1siRNA表达质粒pSilecer K-IE-IgP APE1siRNA与eGFP以脂质体共转染KM3细胞,经G418筛选获得稳定转染的细胞克隆,通过Western blot分析验证APE1siRNA在KM3细胞中的表达.结果 MM特异APE1siRNA表达质粒与eGFP共转染,转染效率约为38%～48%,且APE1siRNA转染细胞较对照组能有效敲除APE1基因表达.结论 APE1siRNA在KM3细胞中获得正确表达.

| Abstract: | Objective To explore the expression of APE1siRNA in KM_3 cells.Methods pSilecer K-IE-IgP APE1siRNA, a multiple myeloma-specific expression vector of APE1siRNA and eGFP plasmid, were transfected into KM_3 cell line by liposome. G418-resistant clones were screened for cell clones with stable transfection. Western blot was used to detect APE1siRNA expression in the transfected KM_3 cells.Results The results of eGFP plasmid suggested that the transfected rate of APE1siRNA plasmid was 38%-48%. Western blot analysis demonstrated that APE1siRNA could knock down the expression of APE1 gene efficiently.Conclusion The expression of APE1siRNA in KM_3 cells may provide the basis for further studies of its biological functions. |
