地塞米松对卵巢癌SKOV-3细胞的影响

摘    要:目的研究地塞米松对体外培养条件下的卵巢癌细胞SKOV-3细胞增殖的影响及诱导其凋亡的情况。方法取对数生长期的SKOV-3细胞(细胞数为5×10^4/mL)分别接种于不同细胞培养板上,试验组每孔加入200μL不同浓度的地塞米松,空白对照组加入等体积PRMI-1640培养液孵育,采用四甲基偶氮唑盐(MTT)比色法检测吸光度(A490值)并计算SKOV-3细胞增殖抑制率;PI染色细胞后上流式细胞仪检测各个时间点的细胞凋亡率。结果MTT法检测结果显示,SKOV-3细胞增殖抑制率随着地塞米松浓度和作用时间的增加均明显增大,以40μmol/L浓度的试验组培养48h的抑制率最高,达(45.25±3.12)%。流式细胞仪检测显示。地塞米松处理后可引起G0/G1期细胞明显增加(P〈0.05),且SKOV-3细胞凋亡率随着地塞米松浓度增加有升高趋势。结论地塞米松能诱导卵巢癌SKOV-3细胞凋亡。

Abstract:Objective To investigate the inhibitive effects of dexamethasone on the proliferation of human ovarian cancer cell line SKOV- 3 and/or its induced apoptosis in vitro. Methods SKOV - 3 ceils (concentration of 5 × 10^4/mL) were harvested in the different cell culture plates. The cells were treated for different time points with different concentrations of dexamethasone. The growth inhibition rates were measured with the MTT method. FCM was used to test the rates of apoptosis and the cell cycle of the cancer cells stained with the fluorescent dye of PI. Results the dexamethasone could inhibit the proliferation of SKOV- 3 cells in a time- dose dependent manner and induce apoptosis of SKOV- 3 cells. The maximum inhibition rate of (45.25 ±3.12)% was detected at 48 h of treatment with 40 μmol/L dexamethasone. Conclusion The cell cycle is significantly stopped at G0/G1 phase by dexamethasone with FCM technology.
