自拟消斑通脉合剂对血管平滑肌细胞增殖与凋亡的影响

[目的]观察中药复方消斑通脉合剂药物血清对体外培养的VSMCs凋亡的影响。[方法]体外培养VSMCs,取对数生长期细胞用于实验,MTT方法观察VSMCs增殖活性;流式细胞仪检测体外培养的VSMCs的凋亡率;透射电镜观察VSMCs形态变化;免疫组织化学方法检测药物血清对VSMCs中P21蛋白表达。[结果]流式法细胞仪显示药物血清可明显诱导VSMCs凋亡,与对照组比较有统计学差异(P<0.01);免疫组化结果提示药物血清可提高VSMCs中P21蛋白表达,与对照组比较有统计学差异(P<0.01)。[结论]中药复方消斑通脉合剂药物血清通过促进VSMCs中P21~(wafl/cipl)蛋白表达而达到抑制VSMCs增殖与诱导凋亡作用。

Effect of Xiao Ban Tong Mai He Ji Drug on Apoptosis in Cultured Vascular Smooth Muscle Cell

[ Objective] To explore the effect of the medical serum of the xiao ban tong mai he ji on Vascular Smooth Muscle Ceils （VSMCs） apoptosis. [ Methods ] VSMCs were treated the medical serum of Xiao Ban Tong Mai He Ji for 24 hours. MTT assay was performed to determine the proliferation of cell. Flow cytometry was applied to detect the apoptosis of VSMCs. Immunohistochemistry was performed to detect the expression of p21 protein. [Results] After the VSMCs were treated with the medical serum, flow eytometry demonstrated that the apoptosis rate of the VSMCs significantly increased. Compared with the control group, the expression of p21 protein was up -grated also. [ Conclusion]The medical serum of Xiao Ban Tong Mai He Ji promotes the apoptosis of VSMCs. These effects are associated with the expression changes of p21 protein.
