腺病毒介导人BMP-7促进去势大鼠椎体骨形成

摘    要:目的探索腺病毒介导人骨形态蛋白-7基因对去势大鼠骨质疏松椎体骨形成情况的影响。方法将去势3个月形成骨质疏松的雌性大鼠的椎体分别按同体对照的方式,不同的椎体分别注入腺病毒介导BMP-7基因、单纯腺病毒载体、及空白椎体对照。术后1周用荧光显微镜观察注射椎体髓腔冲洗液标记荧光的情况以分析椎体内细胞感染腺病毒的情况。术后1月用组织形态学的方法观察椎体骨小梁的情况,骨形态定量学检测骨形成的静态及动态参数,并与未去势大鼠椎体进行比较。结果术后1周注射AdBMP-7的骨质疏松椎体的髓腔冲洗液中可以看到标记荧光。术后1月组织形态学方面,AdBMP-7注射椎体的骨小梁在分布及粗细上与其余各组明显不同,植入AdBMP-7的椎体的骨小梁比较完整,而其他对照椎体骨小梁为明显的骨质疏松的表现,有断裂、变细、盲端等。与无骨质疏松的正常对照组相比,术后1月的AdBMP-7植入椎体的骨小梁不如未去势鼠的椎体。骨形态定量方面的各项指标中,类骨质平均厚度、骨吸收表面、矿化沉积率等参数在AdBMP-7注入组椎体的终板下区中较其他各组无显著差异,平均骨小梁厚度、骨形成表面、骨小梁体积百分数较各对照组椎体增高,各组与未去势大鼠椎体有显著差异,各项动静态指标不如未去势大鼠椎体。结论腺病毒介导人骨形态蛋白-7基因能够促进去势鼠骨质疏松椎体的骨形成。

Abstract:Objective To evaluate the bone formation in the vertebral body of ovariectomized rats by hBMP-7 gene mediated by adenovirus.Methods On the basis of animal model of osteoporosis in the ovariectomized female SD rats and the recloned AdBMP-7,the AdBMP-7 and Ad were injected into the vertebral bodies of osteoporosis rats.One week and 1 month after injection,the fluoroscopy,histology and bone histomorphometry of the injected vertebral bodies were observed.Results By fluoroscopy,1 week after operation,the lavage fluid of injected vertebral body in ovariectomized rats showed fluorescence obviously;by histology,the trabeculae of vertebral bodies injected with AdBMP-7 were lined more density than that of control vertebral bodies.Moreover,the control trabeculae showed sharp and slender,but the trabeculae injected with adBMP-7 not.By the bone histomorphometry,TBV,TFS,TBS and MTPT increased significantly 1 month after the AdBMP-7 were injected.Conclusion The AdBMP-7 may promote the transfection cells in the osteoporosis vertebral body in ovariectomized female SD rats,and enhance the bone formation of the osteoporosis vertebral body.
