摘    要:目的应用蛋白质组学技术筛选人胆汁中与胆管癌相关的蛋白质分子。方法收集2008年3月至2009年10月重庆医科大学附属第一医院肝胆外科经明确病理诊断的胆管癌住院患者的胆汁共15例,正常人胆汁5例。应用非离子型脱脂剂结合分子筛超滤方法制备纯化胆汁样本,通过高效液相色谱-质谱法(HPLC-CHIP-MS/MS)鉴定差异表达的蛋白质,并用Spectrum Mill software对检测结果进行分析。结果与正常人胆汁比较,在胆管癌患者胆汁中,检测到109个高可信蛋白,其中12个为新增蛋白质,其功能主要涉及癌细胞的发生、增殖、分化、转移等。结论高效液相色谱-质谱法鉴定的12个差异表达蛋白质可能成为研究胆管癌的生物学分子标志物。

Abstract:Objective To screen proteins associated with cholangiocarcinoma in human bile with proteomic techniques.Methods Bile specimens were collected from 15 inpatients,who were diagnosed with cholangiocarcinoma in the First Affiliated Hospital of Chongqing Medical University,China,from March 2008 to October 2009,and 5 healthy persons.The bile specimens were purified with nonionic lipid removal reagent CleanasciteTM and ultrafiltration(molecular weight cutoff=10×103).HPLC-CHIP-MS/MS was employed to identify the differentially expressed proteins in the purified bile specimens,and the results were analyzed with Spectrum Mill software.Results Compared with the bile specimens from the healthy persons,109 highly reliable proteins were detected in those from the inpatients with cholangiocarcinoma,12 of which were first reported in proteomic studies of bile.These 12 proteins were mainly associated with oncogenesis,proliferation,differentiation,metastasis,etc.of tumor cells.Conclusion The 12 proteins reported here are potential tumor markers of cholangiocarcinoma.
