摘    要:〔目的〕研究了O1群霍乱弧菌免疫层析试纸的制备方法,通过对试纸材料的处理达到优化胶体金免疫层析体系的目的。〔方法〕建立了快速检测食品中O1群霍乱弧菌的胶体金免疫层析方法,并应用该方法对弧菌属、假单胞菌属及其他常见肠道致病菌,共29株进行检测,同时对180份水产品进行了样品添加试验,并用《出口食品中霍乱弧菌检验方法》(SN/T1022-2001)进行确证,全面评估了该方法的灵敏度、特异性、准确性等。〔结果〕该方法检测食品中O1群霍乱弧菌的灵敏度为105cfu/ml,与食品中常见的致病菌无交叉反应。该方法检测水产品中O1群霍乱弧菌的假阳性率为6.25%。〔结论〕该方法操作简便,灵敏度高,特异性强,准确性高,适用于食品的快速检测。

Abstract:Objective To study the prepareration of colloidal gold dipstick for rapid dtection of Vibrio cholerae O1 serotype,the material optimization of colloidal gold immunochromatography system.Method The method of colloidal gold immunochromatography for rapid detection of Vibrio cholerae O1 serotype in food sample was established.29 strains of different foodborne pathogens,including Vibrios,Psedomonas and other intestinal pathogens were tested with the colloidal gold dipstick.180 pieces of Aquatic Products samples being added Vibrio cholerae O1 serotype were tested with the colloidal gold dipstick and confirmed with SN/T1022-2001.The sensitivity,the specificity and the presision of the method were evaluated.Result The sensitivity of the method for the detection of Vibrio cholerae O1 serotype in Aquatic Products was 105 cfu/ml.There was no cross-reaction with usual intestinal pathogens and the fales-positive rate for the method was 6.25% in Aquatic Products samples testing.Conclusion The rapid detection method provides high sensitivity and specificity,so it can be extensively used to fast screen of plenty of samples.
