谷胱甘肽S-转移酶基因型多态分布与尿细胞学监护结果的关系

摘    要:研究旨在探讨与膀胱癌发生危险有关的谷胱甘肽S-转移酶(GSTs)M1与T1基因型的多态性的分布与有联苯胺接触的膀胱癌高危人群医学监护结果的关系。将上海市染料经工行业的联苯胺接触人群按监护的巴氏分级结果分组,分别与健康对照人群作各种基因型构成比的比较,并进行各组间的构成比趋势分析。结果发现,接触人群内监护史中最大级别≤2级组的T10/0型、M10/0型、和T10/0-M10/0型和2.5级组的M10/0型、T10/0-M10/0型的构成比均显著高于对照人群;平均级别在1.7级以下者中T10/0型和T10/0-M10/0型构成比均显著高于对照人群。另外,在T10/0型和T10/0-M10/0型的构成双上存在着最大级别越 低构成比越高的显著趋势。结果提示,GSTs基因检测与膀胱脱落细胞检查可能是两种具有不同意义的指标,前者反映的是易感性,后者则可认为是临床前期诊断指标,两者之间不存在相关关系是可能的。但本研究中出现的负相关关系是否反映GSTM1和T10/0基因型对膀胱上皮细胞异变的逆转具有一定的保护作用,值得进一步研究。

Abstract:This study was aimed to ascertain the relationship between the distribution of glutathione S-transferase(GSTs)M1 and T1 gene polymorphism related to the ris k of bladder cancer development and the medical surveillance for the bladder cancer high risk population exposed to benzidine.The population occupationally exposed to benzidine in Shanghai dyestuff industry was divided into several groups based on the surveillance results with Papanicoloaus method,and the proportions of genotypes in each group were compared with that in the control population.The trend analyses was made for the inter-group proportions.The results showed that the proportions of T1 0/0,M1 0/0,and T1 0/0-M1 0/0 in the group of ≤2,and M1 0/0 and T1 0/0-M1 0/0 in the group of 2.5 of the highest grade during the surveillance were higher than those in the control population.The proportions of T1 0/0and T1 0/0-M1 0/0 in the group of 1.7 of the average grade du ring the surveillance were higher than that in the control population.Moreover,th ere was a significant trend of the lower highest grade with the higher proportion in T1 0/0 and T1 0/0-M1 0/0.The results suggested that the detection for GSTs genotype may be the index different to the urine exfoliation examination.The former reflected the susceptibility,and the latter should be regarded as the diagnostic index for pre-clinic.Maybe,there is no correlation between them.However,whether the fact of the negative correlation presented in this study reflects GSTs M1 and T1 genotype 0/0 may protect the reversion from the variation of bladder epithelia,will be valuable for further research.
