丙泊酚对大鼠急性肾缺血再灌注损伤的抗氧化机制研究

摘    要:目的:探讨丙泊酚预处理对急性肾缺血再灌注损伤(acute renal ischemia reperfusion injury,ARIRI)的抗氧化保护作用及其机制。方法:建立大鼠急性肾缺血再灌注损伤模型,健康雄性SD大鼠54只,随机分为3组:假手术组、缺血再灌注(ischemia/reperfusion,I/R)组、丙泊酚预处理组。缺血再灌注前15min、缺血再灌注后2h、24h分别检测血清尿素氮(BUN)、肌酐(Cr)、超氧化物歧化酶(SOD)、丙二醛(MDA)及观察肾组织的病理学变化。结果:丙泊酚预处理组肾组织病理学变化明显轻于缺血再灌注组。与假手术组比较,缺血再灌注组中血清尿素氮、肌酐水平显著增加(P<0.05),而丙泊酚预处理组尿素氮、肌酐水平则低于缺血再灌注组(P<0.05)。丙泊酚预处理组血清超氧化物歧化酶和丙二醛水平与缺血再灌注组均有统计学差异(P<0.05)。结论:在急性肾缺血再灌注损伤中,丙泊酚能起到对肾脏的保护作用。

Abstract:Objective To investigate the renal protection and mechanism of propofol in ARIRI rat models. Methods The model of rat's ARIRI was established. 54 SD rats were randomly divided into 3 groups: false operating group, ischemia reperfusion group and propofol group. BUN, Cr, SOD, and MDA were detected before I/R for 15 minutes and after I/R for 2 hours and 24 hours, and renal histopathology lesions were examined.Results Renal histopathology lesions of propofol preconditioning group decreased significantly after I/R. In I/R group, BUN and Cr serum levels increased significantly after I/R, compared with false operating group(P<0.05), while propofol decreased the serum levels of BUN and Cr significantly(P<0.05).There was statistic significance of the serum levels of SOD and MDA in Propofol preconditioning group and I/R group(P<0.05).Conclusion Propofol preconditioning to kidney could protect the kidney from ARIRI.
