摘    要:目的构建表达红荧光(DsRed)重组腺相关病毒,研究腺相关病毒对骨髓间充质干细胞(BMSCs)的转染效率。方法DsRed目的基因经酶切插入载体质粒pAAV-MCS,构建重组质粒pAAV-DsRed。磷酸钙法转染AAV-293细胞获取重组腺相关病毒。将AAV-DsRed体外转染原代培养的兔BMSCs,分析其转染效率。结果成功构建重组质粒pAAV-DsRed,获得腺相关病毒AAV-DsRed,并感染兔BMSCs。结论腺相关病毒能够转染BMSCs并表达外源基因,具有应用于临床治疗的潜力。

Abstract:Objective To construct the recombinant adeno-associated virus expressing red fluorescent(DsRed)protein and investigate its transfection efficiency in vitro.Method The recombinant plasmid pAAV-DsRed was constructed by inserting the DsRed gene into the pAAV-MCS and generating the primary recombinant AAV by calcium phosphate-based technology. The transfection efficiency were evaluated by transfecting AAV-DsRed into primary rabbit BMSCs.The direction of the insertion was confirmed by PCR analysis. Results The recombinant plasmid pAAV-DsRed has been constructed successfully, and AAV-DsRed obtained;the primary BMSCs could be transfected efficiently with AAV-DsRed. Conclusion The recombinant Adeno-associated virus was constructed succeesfuly,it can transfect BMSCs and express exogenous gene, thus possess the potential for clinical application.
