复发性生殖器疱疹患者外周血T细胞亚群及Th1/Th2型细胞因子的模式

目的分析复发性生殖器疱疹 ( RGH)患者外周血中 T细胞亚群及 Th1/Th2型细胞因子网络的偏移及其在发病机理中的作用。方法采用三色和双色荧光抗体染色技术经流式细胞仪检测 2 0例 RG H患者和正常对照组外周血 T细胞亚群及 CD4 + T、CD8+ T细胞内 4种细胞因子染色阳性细胞百分率。结果 RGH患者 CD3+ T细胞、CD4 + T细胞、CD4 + T/CD8+ T明显低于正常对照水平 ( P<0 .0 0 1) ;IL- 2、IFN- r、IL- 12阳性细胞百分率均显著降低 (分别为 CD4 + T:P<0 .0 0 1,CD8+ T:P<0 .0 5) ;IL- 4阳性细胞则明显升高 ( CD4 + T:P<0 .0 5,CD8+ T:P<0 .0 0 1)。结论 RGH患者存在 T细胞亚群和 Th1、Th2细胞分泌细胞因子交互作用失衡 ,后者表现为 Th2型细胞因子( IL - 4)产生过多 ,而 Th1型细胞因子 ( IL - 2、IL - 12、IFN - r)产生过少 ,Th1/Th2失衡在 RGH的发病机制中可能具有重要的作用。

Study on the T Lymphocyte Subsets and Th1/Th2 Cytokine Profile in Peripheral Blood of Patients with Recurrent Genital Herpes

Objective To analyze the T lymphocyte subsets and the shift of Th1/Th2 type cytokine profile in peripheral blood of patients with recurrent genital herpes (RGH) and its role in the pathogenesis of RGH.Methods Three color and two color immunofluorescent staining of cell surface antigen and intracellular cytokines(IL-2?IL-4?IL-12?IFN-r)in peripheral blood from 20 patients with RGH were performed and then analysed by flow cytometry.Results Compared to controls,in patients with RGH CD3 +T,CD4 +T and CD4 +T/ CD8 +T cells ratio were significantly decreased (P<0.001),and IL-2,IFN-r and IL-12 producing T cells were increased(CD4 +T:P<0.001,CD8 +T:P<0.05 respectively),whereas IL-4 producing T cells were increased(CD4 +T:P<0.05;CD8 +T:P<0.001 respectively).Conclusions RGH patients seem to have lost the balance between T lymphocyte subsets and Th1/Th2 cytokines.The Th2 cells have increased and predominated.The off balance Th1/Th2 may play an important role for the pathogenesis of RGH.
