HPLC法测定复方利血平片中氢氯噻嗪、盐酸异丙嗪和利血平的含量

目的 建立HPLC法测定复方利血平片中氢氯噻嗪、盐酸异丙嗪和利血平的含量。方法采用C18柱为固定相（150mm×4．6mm×5μm），0．01mol·L^-1庚烷磺酸钠：乙腈：三乙胺（53：47：0．1），用磷酸溶液调节pH至5．5为流动相，检测波长为230～380nm。结果氢氯噻嗪在0．1934—2．9013g·L^-1浓度范围内线性关系良好，r=0．9999，平均加样回收率为100．44％，RSD为0．54％（n=6）；盐酸异丙嗪在0．135～2．025g·L^-1浓度范围内线性关系良好，r=1．0000，平均加样回收率为100．25％，RSD为0．18％（n=6）；利血平在1．965—29．475mg·L^-1浓度范围内线性关系良好，r=1．0000，平均加样回收率为100．73％，RSD为0．47％（n=6）。结论该方法具有操作简便、结果准确的优点，可用于复方利血平片中氢氯噻嗪、盐酸异丙嗪和利血平的含量。

Determination of hydrochlorothiazide,promethazine hydrochloride and reserpine in compound reserpine tablets by HPLC

Aim To establish a method for determination of hydrochlorothiazide,promethazine hydrochloride and reserpine in compound reserpine tablets by HPLC.Method C18 column(150 mm×4.6 mm×5 μm)was used,the mobile phases consisting of 0.01 mol·L-1 sodium heptanesulfonate solution-acetonitrile-triethylamine(53∶47∶0.1).The detection wavelength was set at 230~380 nm.Results The linear range of hydrochlorothiazide was 0.1934~2.9013 g·L-1.The average recovery was 100.44%,with of 0.9999,and RSD of 0.54%(n=6).The linear range of promethazine hydrochloride was 0.135~2.025 g·L-1.The average recovery was 100.25%,with r of 1.0000,and RSD of 0.18%(n=6).The linear range of Reserpine was 1.965~29.475 mg·L-1.The average recovery was 100.73%,with r of 1.0000,and RSD of 0.47%(n=6).Conclusions The method was accurate.It could be used for determination of hydrochlorothiazide,promethazine hydrochloride and reserpine in compound reserpine tablets.
