奥沙利铂对 K562细胞体外生长的影响

摘    要:目的:观察奥沙利铂对人白血病K562细胞的作用.方法:检测奥沙利铂对K562细胞作用的时间效应和剂量效应,在电镜下观察奥沙利铂作用后细胞形态变化;用流式细胞仪分析细胞周期的改变.结果:随着作用时间及药物浓度的增加,奥沙利铂对K562细胞的增殖抑制作用就越明显.0.016mmol/L奥沙利铂作用72h,K562细胞生长抑制率达85%,形态学及流式细胞仪分析显示G2/M期阻滞和细胞凋亡.结论:奥沙利铂诱发的人白血病细胞的凋亡与细胞分裂期阻滞密切相关.

Abstract:Objective:To investigate the effects of oxaliplatin on human leukemia K562 cell and its mechanism.Methods:The leukemia cell growth was measured by MTT-microculture tetrazolium assay.Cell-cycle kinetics and apoptosis were analyzed by flow cytometry and microscopic examination.Results:Oxaliplatin inhibited the leukemia cell growth in concentration- and time-dependent manners.Marked cells accumulated in G_2/M phase.The rate of cell growth inhibition was 85% after intervention in 0.016 mmol/L oxaliplatin for 72 hours.The cells showed G_2/M arrest and apoptosis.Conclusion:Oxaliplatin suppresses the growth of K562 cells in vitro by causing cell-cycle arrest,and cell apoptosis.The mechanisms of oxaliplatin provide theoretical guidance for clinical treatment.
