| Abstract: | Objective To establish a HPLC method for the determination of norvancomycin concentration in human serum.Methods The Shima-pak C18 column（150 mm×4.6 mm,5 μm） was used with a mobile phase of methanol/potassium dihydrogen phosphate buffer（15∶85,pH 3.2）.The detection wavelength,flow rate and column temperature were set at 236 nm,1 mL·min-1 and 35 ℃,respectively.Norvancomycin was used as the internal standard.Results The retention time of norvancomycin and vancomycin was 5.8 minutes and 7.1 minutes,respectively.The regression equation of norvancomycin was：Y=（0.000 203 27）X-1.624 06,r=0.999 2.The calibration curves obtained were linear in the range of 2.0~100.0 mg·L-1.Conclusion The HPLC method is convenient,sensitive,accurate and reproducible for determining the concentrations of norvancomycin in human serum and investigating its pharmacokinetics. |
