瘦素受体基因外显子6 Gln223Arg变异与高血压左心室功能及肾脏血流动力学的关系

摘    要:目的:探讨原发性高血压患者的左心室功能、肾脏血流动力学指标与瘦素受体(LR)基因Gln223Arg变异的关系。方法:用聚合酶链反应—限制性片段长度多态性(PCR-RFLP)检测了原发性高血压患者90例(高血压组)及对照组52例的LR基因Gln223Arg的变异,对两组的LR基因Gln223Arg的变异进行对照分析。同时应用彩色多普勒超声仪检测了高血压组及对照组的左心室功能、肾脏血流动力学指标,并进行对照分析。结果:LR基因Gln223Arg变异的不同基因型在高血压组与对照组之间分布频率无显著差异;高血压组的左心室功能、肾脏血流动力学指标与LR基因Gln223Arg不同基因型相关,GG基因型高血压患者的左心室功能、肾脏血流动力学各项指标变化更为明显(P<0.05或<0.01);对照组的肾脏血流动力学指标(尤其是阻力指数)与LR基因Gln223Arg不同基因型相关(P<0.05)。结论:原发性高血压患者的左心室功能、肾脏血流动力学指标的变化与LR基因Gln223Arg的变异有关。

Abstract:Objective:To investigates if the leptin receptor gene polymorphism is associated with essential hypertension.To investigate if the leptin receptor polymorphism is associated with the renal hemodynamics and left ventricular function.Methods:Polymerase chain reaction restriction fragment length polymorphisms(PCR-RELP)technique was used to detect the leptin receptor gene polymorphism.The color Doppler echocardiography was used to detect the left ventricular function and the renal hemodynamics.The study covered 90 hypertensive and 52 controls.Results:The frequencies of leptin receptor genotypes in hypertensive and that in control was no significant difference.The leptin receptor gene polymorphism is associated with renal hemodynamics and the left ventricular function The hypertension with GG genotype was significantly different in left ventricular function and renal hemodynamics indexes to normal controls and hypertension with AA or GA genotype(P<0.05).Conclusion:The renal hemodynamics and the left ventricular function was associated with leptin receptor gene polymorphism.
