阿霉素中毒大鼠背根神经节神经元及细胞核孔的改变

摘    要:目的 探讨阿霉素中毒所致神经元坏死过程中胞核,胞质及核孔的变化,是否存在凋亡。方法 SD大鼠静脉注射阿霉素8mg/kg,使其腰背根神经节细胞发生变性。利用光镜,电镜、聚焦激光扫描显微镜,TUNEL染色等方法观察不同时期神经元和核孔的变化,寻找凋亡存在的证据。结果 阿霉素应用14d后,核膜切线位核孔的结构变得模糊不清,数量减少;垂直位核膜隔明显稀疏,甚至消失,神经核中度变性及胞质明显变性。结论 核孔

Abstract:Objective To observe the changes of the nucleus,cytoplasm and nuclear pore during doxorubicin (DXR) produced degeneration of neurons in the lumbar dorsal root ganglion (DRG) in rats. Methods The changes of the DRG neuron and nuclear pore of Sprague Dawley rats and the presence of apoptosis were studied by using light and electronic microscopy and terminal deoxynucleotidyl transferase mediated deoxyuridine triphosphate biotin nick end labeling method in varying time during 1 to 20 days after intravenous 8 mg/kg of DXR was administered. Results Observation revealed the degeneration of the moderate nuclear and remarkable cytoplasm. In some neurons, there appeared mostly dark and usually moderate degrees of nuclear degenerative changes, and the nuclear pores were decreased in number and becoming obscured 14 and 20 days after DXR administration. As DXR enters presumably the nucleus is partly removed through the nuclear pores. However, the diameters of nuclear pores were similar to the DXR intoxicated and control rats. Conclusion The changes in nuclear pores of neurons in DXR intoxication are considered to be part of the degenerative or necrotic changes of DRG neuron and there appears no apoptosis.
