软组织肉瘤中p53基因的突变和蛋白的表达研究

目的检测软组织肉瘤中p53基因的突变和蛋白的表达情况。方法应用PCR-SSCP和DNA测序的方法检测p53基因的突变,免疫组织化学技术Envision二步法检测p53蛋白的表达。结果 63例软组织肉瘤中检测到p53基因突变18例（28.6%）,突变位点主要在第6外显子和第7外显子,突变和组织学分级间无明显相关性;63例p53蛋白的表达44例,蛋白的表达与组织学分级间无明显相关性;且p53基因的突变与p53蛋白的表达无明显相关性。结论 p53基因的突变和p53蛋白的表达可能作为软组织肉瘤发生的一个重要因素。

p53 gene mutations and p53 protein expression in soft tissue sarcomas

Objective To investigate the expression of p53 gene mutations and p53 protein expression in soft tissue sarcomas.Methods Tissue specimens were collected from 63 cases of soft tissue sarcomas.p53 gene mutation and p53 protein expression were detected by single strand conformational polymorphism（SSCP）,DNA sequencing and immunohistochemical staining,respectively.Results Mutations of p53 gene were detected in 18（28.6%） of 63 cases of soft tissue sarcomas.Most of the mutations were point mutation in exon 6-7,there was no significant correlation between p53 mutation and histological grade.Expression of p53 protein was detected in 44 cases,there was no significant correlation between p53 protein expression and the histological grade;and the p53 gene mutations were not correlated with the expression of p53 protein either.Conclusion p53 gene mutations and p53 protein expression may be involved in oncogenesis of soft tissue sarcomas.
