氧化苦参碱肠内菌代谢及吸收入血活性成分的研究

摘    要:目的 :通过离体和整体实验观察人肠内菌对氧化苦参碱的代谢 ,并确定吸收入血的代谢物。方法 :通过薄层色谱和高效液相色谱仪定性检测氧化苦参碱及其代谢产物。并通过UV ,IR ,NMR及MS等光谱学方法确定代谢物的结构。结果 :离体实验结果表明 ,氧化苦参碱可被人肠内菌代谢成苦参碱。给大鼠按 100mg·kg-1灌胃氧化苦参碱 3h后 ,剪头处死取血 ,以TLC和HPLC检测血清中代谢产物。证明氧化苦参碱和苦参碱均能被吸收入血。结论 :人肠内菌可将氧化苦参碱代谢成苦参碱 ,上述两种生物碱均可被吸收入血。

Abstract:Objective : To study the oxymatrine metabolism induced by human intestinal bacteria and its absorbed metabolites in blood. Method :TLC and HPLC were used to examine oxymatrine and its metabolites, and UV, IR, NMR and MS were used to confirm the chemical structures of the metabolites. Result: Oxymatrine was transformed into matrine by human intestinal bacteria metabolism in vitro. Rats were given orally oxymatrine 100 mg·kg-1 and were decapitated 3 hours after administration, and their blood was taken to examine serum metabolites by TLC and HPLC, which revealed that oxymatrine and matrine were absorbed into blood. Conclusion:Oxymatrine will be transformed into matrine when it is given orally and both of the alkaloids can be absorbed to blood.
