摘    要:为了观察重组人骨形态发生蛋白-7(rhBMP-7)基因转染骨髓基质细胞(BMSCs)后的表达及表达产物对BM-SCs增殖的影响,用脂质体介导法将rhBMP-7基因转染BMSCs,用逆转录PCR技术和免疫组化SABC法分别检测其瞬时表达和稳定表达;再用^3H-TdR掺入法检测基因表达产物对正常培养的BMSCs增殖的影响。结果表明:转基因细胞能高效表达外源基因,且表达时间长达4周;基因表达产物能明显促进BMSCs的增殖。该结果为基因增强的组织工程技术用于关节软骨缺损的修复提供了理论依据。

Abstract:In order to investigate the expression of recombinant human bone morphogenetic protein 7 (rhBMP 7) gene in transduced bone marrow stromal cells (BMSCs) and the bioactivity of transgene expression products, BMSCs were transfected with rhBMP 7 gene via lipofectamine vector. The transient and stable expression of the transgene was detected by using RT PCR assay and immunohistochemical SABC method respectively. The influence of the exogenous gene products on BMSCs cultured in normal conditions was analyzed by 3 H TdR incorporation method. The results showed that the exogenous gene could be expressed by the transduced BMSCs effectively and the duration was up to 4 weeks. The transgene expression products could stimulate the proliferation of BMSCs cultured in normal conditions. It is suggested that it is possible to use gene enhanced tissue engineering technology for the repair of articular cartilage defects.
