摘    要:目的观察目的基因在晶状体上皮细胞的表达情况,以探讨基因疗法用于后发性白内障的可行性。方法用绿色荧光蛋白基因作为报告基因,以逆转录病毒载体携之转染晶状体上皮细胞,观察GFP在晶状体上皮细胞的表达情况。结果基因转染后可见晶状体上皮细胞的GFP表达率在30%~35%。结论逆转录病毒载体可以携带目的基因转染于晶状体上皮细胞,并达到较高的转染率。

Abstract:Object iv e To study the possibility of prevention of posterior capsular opacification(PC O)by transfer of exogenous gene into lens epithelial cells.Methods Green fluo rescence protein(GFP)gene was used as a reporter gene,and was transferred int o lens epithelial cells mediated by retroviral vector,GFP expression in cells w as analyzed.Results The positive GFP gene was about 30%-35%in lens epith elial cells.Conclusion Retrovirus-mediated gene may be the therapeutically val uable for the treatment of posterior capsular opacification.
