炎调方对脓毒症大鼠模型炎症介质的影响

摘    要:目的观察脓毒症大鼠血清TNF-α、IL-6、IL-10的释放及血管活性肠肽(VIP)的改变,旨在研究炎调方有效治疗脓毒症的作用机理。方法采用盲肠结扎穿孔术制备脓毒症大鼠模型,将雄性SD大鼠随机分为正常组、假手术组、模型组、炎调方组、减方对照组(炎调方去大黄、芒硝),分别于造模后12、24、36h处死动物,双抗体夹心酶联免疫吸附法测定血清TNF-α、IL-6、IL-10、VIP水平。结果与正常组相比。假手术组大鼠血清TNF—d、IL-6、VIP水平相当(P〉0.05);模型组12h的血清TNF—α、IL-6、VIP水平和24h的血清TNF—α、IL-6水平均高于正常组(P〈0.叭),模型组24h血清VIP水平则与正常组相当(P〉0.05);与模型组相比,炎调方组造模后12h血清TNF—d、IL-6、VIP水平即有明显下降(P〈0.05或0.01),但在24h时差异无统计学意义(P〉0.05);减方对照组(炎调方去大黄、芒硝)和模型组比较12、24h均无统计学意义(P〉0.05);与减方对照组相比,炎调方组血清TNF—α、IL-6、VIP水平在12h水平明显下降(P〈0.05或0.01),但在24h则差异无统计学意义(JP〉0.05);模型组、炎调方组和减方对照组的IL-10随着时间延长而逐渐下降。炎调方组和模型组、减方对照组相比,IL-10在12h明显升高(P〈0.05),但在24h内差异无统计学意义(P〉0.05)。结论炎调方能明显下调脓毒症大鼠血清TNF—α、IL-6、VIP水平,对IL—10有-定上调作用。

Abstract:Objective: To observe the levels of TNF—α,IL-6,IL-10 release and vasoactive intestinal peptide changes, and to study the mechanism of Yantiao Decoction in the treatment of sepsis. Method: Sepsis rat model was induced by cecal ligation and puncture sepsis rat model. Male SD rats were randomly divided into Normal group, Sham operation group, Model group, Yantiaofang group ,The minus side in the control group (Yantiaofang group Minus the rhubarb, Glauber's salt),were killed after modeling 12 h,24 h, 36 h respectively, by ELISA mea- sure serum of TNF—α, IL-6,IL-10,VIP level. Results: Compared with the normal group, sham operation group rates, serum of the the IL-6, the TNF—α and VIP level showed no significant difference(P〉 0.05 );Made mode after 12 h and 24 h,the model group was significantly higher than normal group serum level of TNF—α,IL-6,VIP(P 〈 0.01 ), but serum level of VIP showed no statistical difference in making mode after 24 h (P〉 0.05 );Compared with the mode group, Yantiao Decoction group, s serum of TNF—α, IL-6, VIP level was lower significantly in mak- ing mode after 12 h (P〈0.05 or 0.01),but there 'have no significant difference in making mode after 24 h (P〉 0.05);Minus side the control group (Yantiaofang group Minus the rhubarb, Glauber's salt) and model group had not all statistical significance in 12 h and 24 h (P〉 0.05 );Compared with the minus side in the control group, the srum TNF—α, IL-6, VIP level of Yantiaofang group is failer significantly in 12 h(P〈 0.05 or 0.01 ), but there have no significant difference in making mode after 24 h (P〉 0.05); The srum IL-10 level of the model group,the Yantiaofang group and the minus side in the control group, was gradually decreased with time lengthen. Compared with model group and the minus side in the control group,the srum IL-10 level of the yantiaofang were signify- cantly increased at 12 h (P〈0.05),but they had not all statistical significance in 24 h (P〉0.05). Conclusion: Yantiao Decoction group can markedly increase the serum level of the IL-10 and decrease the serum levels of the TNF—α, IL-6,and VIP of the septic rats.
