哮喘豚鼠下呼吸道及内脏传入部位NGF表达的研究

摘    要:目的 探讨哮喘豚鼠下呼吸道和内脏传入系统神经生长因子 (NGF)的作用。 方法 利用免疫组织化学和原位杂交结合显微图像分析 ,研究哮喘豚鼠下呼吸道和内脏传入系统NGF免疫反应及mRNA表达的变化。 结果 与对照组相比 ,哮喘豚鼠NGF免疫反应在气道上皮、C7~T5段脊神经节及脊髓背角明显上调 (P <0 0 1) ,NGFmRNA表达在气道上皮明显增多 (P <0 0 1) ,而在C7~T5段脊神经节及脊髓背角却未见明显改变。 结论 气道上皮、C7~T5段脊神经节及对应节段脊髓背角的NGF可能参与哮喘的发病过程 ;C7~T5段脊神经节细胞及对应节段脊髓背角神经细胞本身能合成NGF。

Abstract:Objective To explore the effect of nerve growth factor(NGF) in the lower respiratory tract and the visceral afferent system of asthmatic guinea pigs. Methods By means of immunohistochemistry and in situ hybridization combined with the micro-image analysis to investigate the alterations of NGF immunoreactivity and mRNA expression in asthmatic guinea pig. Results The positive NGF immunoreactivity was increased in airway epithelia,C-7-T-5 spinal ganglia and correspondent spinal dorsal horn in the asthmatic guinea pigs compared with the controls(P<0.01).The NGF mRNA expression was also upregulated in the airway epithelia,but not in C-7-T-5 spinal ganglia and correspondent spinal dorsal horn in the asthmatic guinea pigs compared with the controls(P<0.01).Conclusions 1.Nerve growth factor in the airway epithelia,C-7-T-5 spinal ganglia and correspondent spinal dorsal horn might be involved in the pathogenesis of the asthma.2.Neurons in the C-7-T-5 spinal ganglia and correspondent spinal dorsal horn can synthesize NGF.
