| 摘 要： | 目的探讨PTEN基因在三株人乳腺癌细胞中的表达及意义。方法采用RT-PCR及Western blot法检测三株人乳腺癌细胞株内源性PTEN mRNA及其产物蛋白的表达。结果PTEN基因的产物蛋白在乳腺癌细胞MCF-7中出现高表达,而在两种恶性乳腺癌细胞中却明显出现低表达或缺失表达。结论PTEN作为一种新型的抑癌基因,其突变与缺失可能与恶性乳腺肿瘤的发生发展密切相关。

| Abstract: | Objective To investigate the significance and expression of PTEN gene in three breast cancer cell lines. Methods RT-PCR was used to detect the expression of the endogenous PTEN mRNA, while Western bloting was used to detect the expression of the endogenous PTEN protein in three breast cancer cell lines. Results The PTEN gene product protein appeared a high expression in the benign breast cancer cell,while it was low expression or the loss expression in two kinds of malignant breast cancer cell. Conclusion As a new tumor suppressor gene,PTEN are related to the occurrence and development of the malignant breast cancer. |
