耐力训练对衰老小鼠心肌Akt/mTOR信号通路的影响

摘    要:目的:探讨耐力训练对衰老小鼠心肌Akt/mTOR信号通路的影响。方法:雄性2月龄SAMP8小鼠随机分为年轻对照组(n=12)、耐力训练组(n=12)和老年对照组(n=12),年轻对照组在3月龄时处死,老年对照组常规饲养至6月龄时处死。耐力训练组从3月龄开始进行3个月的跑台耐力训练至6月龄时处死,每周3次,每次40min。测定各组小鼠心肌重量并计算心系数,取心肌组织进行HE染色观察;使用实时荧光定量PCR技术测定心肌Akt/mTOR信号通路中Akt、mTOR、p70S6K和eIF-4E mRNA的表达。结果:耐力训练组小鼠心脏重量和心系数与老年对照组相比显著增加(P<0.05),组织学检测未发现其心肌细胞出现病理性改变。老年对照组小鼠心肌Akt、mTOR、p70S6K和eIF-4E mRNA表达与年轻对照组相比显著下降(P<0.05);耐力训练组心肌各靶物质mRNA表达较老年对照组显著增加(P<0.05),低于年轻对照组,但不具有统计学意义。结论:耐力训练可以通过提高心脏Akt/mTOR通路中各信号分子的表达,使衰老心脏发生生理性的运动性心肌肥大。
