川红拔瘤膏对小鼠脑胶质瘤生长的影响及作用机制

目的:观察川红拔瘤膏外敷对小鼠G422脑胶质瘤生长的抑制作用及其抗肿瘤的机制。方法:选取清洁级小鼠50只,随机分为:对照组、环磷酰胺组、川红拔瘤膏高、中、低3个剂量组。中药组右腋下皮肤涂抹给药,给药量为0.5mL。连续给药14天,阳性对照组连续给药7天。连续观察川红拔瘤膏对肿瘤生长的抑制情况,末次处死后,取肿瘤组织,流式细胞仪检测细胞周期变化与细胞凋亡。RT-PCR法检测肿瘤组织中caspase-3的表达。结果:拔瘤膏高浓度组的瘤重(0.516±0.107)g与其他组比较(除环磷酰胺组)对实体瘤的生长具有显著的抑制作用(P0.05)。高浓度组肿瘤抑制率为31.28%。流式细胞仪结果显示,与空白组比较川红拔瘤膏高剂量组明显将小鼠肿瘤组织细胞阻滞于G0/G1期(P0.05),S期细胞数明显减少(P0.05)。细胞凋亡分析可知,高剂量组作用的肿瘤细胞的凋亡率为(23.6±4.35)%,明显高于对照组(8.86±1.23)%,差异有统计学意义(P0.05)。其他组与对照组比较,差异无统计学意义。caspase-3分析结果显示,与低剂量组、中剂量组,caspase-3表达强度无明显改变(8.23±0.43),(7.96±0.52)vs(7.24±0.36),P0.05]。高剂量组的caspase-3表达显著增加(23.45±2.31)vs(7.24±0.36),P0.01]。结论:川红拔瘤膏对小鼠G422脑胶质瘤具有抑制生长的作用,其作用可能与其抑制肿瘤细胞DNA合成、周期阻滞、诱导凋亡及上调凋亡基因caspase-3表达有关。
