摘    要:目的观察Sina基因同源类似基因1(SIAH1)在大鼠脊髓损伤过程中的表达情况,分析它们与神经元存活和死亡、胶质细胞活化增殖等生物学的关系,探讨它们在脊髓损伤和修复过程中的生物学功能。方法制作脊髓损伤模型,利用Western blot、免疫组织化学、免疫荧光技术观察SIAH1在损伤脊髓中的表达变化、组织分布与细胞定位,在此基础上探讨SIAH1在脊髓损伤后的病理生理意义。结果 Western blot结果显示SIAH1经历了在损伤后8h、1d和2 d的增加后开始下降,直到损伤后14 d仍没有明显上凋的变化趋势。免疫组化结果显示在脊髓损伤后1d,SIA H1的阳性信号在临近损伤中心的脊髓中明显增加,而在脊髓损伤后7 d,SIAH1阳性的细胞数明显减少。免疫荧光结果显示SIAH1与神经元、星形胶质细胞和小胶质细胞存在共定位。结论脊髓损伤后脊髓中SIAH1的表达上调可能参与神经元的凋亡过程以及星形胶质细胞和小胶质细胞的活化的病理生理过程。

Abstract:Objective To observe the expression patterns of absentia homolog 1(SIAH1) in rat models of spinal cord injury,and the association with neuronal survival or apoptosis and glial proliferation was analyzed.As well as to investigate their potential biological roles in nerve injury and regeneration. Methods Sprague-Dawley(SD) rats were subjected to spinal cord injury.In the purpose of exploring the pathological and physiological significance of SIAH1,the expression patterns and distribution for SIAH1 were observed by Western blot,and tests of immunohistochemistry and immunofluorescence.Results Western blot analysis showed SIAH1 protein level was significant up-regulated at the 8th hour,1st day and 2nd day after injuried,then reduced and there was no up-regulation till the 14th day.Immunohisto-chemical analysis showed that the immunochemical staining was increased at the 1st day and reduced at the 7th day after injury.Immunoflurescence analysis showed SIAH1 were expressed in neurons,astrocytes and microglias.Conclusion The up-regulation of SIAH1 in the injured spinal cord may associate with apoptosis process of neurons and glial proliferation.
