蛇床子素对小鼠酒精性脂肪肝的治疗作用

目的观察蛇床子素对小鼠酒精性脂肪肝的治疗作用并初步探讨其作用机制。方法建立小鼠酒精性脂肪肝模型后,分别以蛇床子素不同剂量（10～40 mg/kg）给予治疗6周,然而测定小鼠血脂、肝脂及肝组织中超氧化物歧化酶（SOD）、丙二醛（MDA）和谷胱甘肽过氧化物酶（GSH-PX）含量,同时观察肝脏的病理变化。结果经蛇床子素处理过的小鼠血清中总胆固醇（TC）、甘油三酯（TG）、低密度脂蛋白-胆固醇（LDL-C）水平和肝组织中TC、TG和MDA的含量明显低于模型组（P〈0.05或0.01）,同时肝组织中的SOD含量明显高于模型组（P〈0.05或0.01）。肝组织形态学评估表明,蛇床子素显著减少了肝脂质的沉积。结论蛇床子素对小鼠酒精性脂肪肝有较好的治疗作用,其作用机制可能与其增加机体的抗氧化能力有关。

Therapeutic Effect of Osthole on Alcohol-Indced Fatty Liver in Mice

Objective To study the therapeutic effect of osthole on alcohol-indced fatty liver in mice and investigate its potential mechanism.Methods A mouse alcoholic fatty liver model was made by feeding alcohol for 6 weeks.Then osthole was administered to these mice in a dose of 10,20 and 40 mg/kg for 6 weeks,the contents of serum and hepatic lipids,the levels of SOD,MDA,GSH-PX in hepatic tissues were measured,the hepatic histological changes were observed.Results The levels of serum total cholesterol（TC）,triglyceride（TG）,low density lipoprotein-cholesterol（LDL-C）,and hepatic tissue contents of TC,TG and malondialdehyde（MDA） in osthole-treated groups were significantly decreased, while the level of superoxide dismutase（SOD） was significantly increased as compared with that in the model group.Moreover,the histological evaluation of liver demonstrated that osthole dramatically decreased lipid accumulation.Conclusion Osthole is effective in treating mouse alcoholic fatty liver,and its main mechanisms might be related to reduction of hepatic oxidative stress.
