胆囊癌组织hPTTG1与c-Myc的表达及意义

摘    要:目的: 探讨人垂体瘤转化基因1蛋白(hPTTG1)和c-Myc在胆囊癌组织中的表达及与其生物学行为之间的关系;并探讨二者的相关性及可能机制. 方法: 用免疫组织化学SP法对41例胆囊癌和22例慢性胆囊炎组织中hPTTG1和c-Myc的表达进行检测. 结果: 41例原发胆囊癌中hPTTG1和c-Myc的表达阳性率分别为82.9%和63.4%,均高于慢性胆囊炎组(P=0.002, 0.002). hPTTG1的表达与临床分期和淋巴结转移有关(P=0.025, 0.007),而与组织学分级无显著关系(P=0.114); c-Myc的表达与淋巴结转移有关(P=0.032),而与组织学分级、临床分期无显著关系(P=0.127, 0.198); hPTTG1的表达与c-Myc的表达呈正相关(r=0. 463, P=0.002). 二者的表达与患者的性别、年龄、肿瘤的种类,是否伴有胆囊结石均无关. 结论: hPTTG1和c-Myc可能在胆囊癌的发生、发展过程中起重要作用,可能成为反映胆囊癌侵袭性的生物学指标,为其治疗提供了新的靶点.

Expression of hPTTG1 and c-Myc in gallbladder carcinoma
Abstract:AIM: To investigate the expression of hPTTG1 and c Myc in human carcinoma of gallbladder and their correlation with clinicobiological behavior and to probe the possible mechanism of their association. METHODS: The expressions of hPTTG1 and c Myc in 41 cases of human gallbladder carcinoma and 22 cases of chronic cholecystitis were detected with immunohistochemistry by SP. RESULTS: The positive rates of hPTTG1 and c Myc were 82.9% and 63.4% respectively, which were significantly higher in gallbladder carcinoma than in chronic cholecystitis ( P = 0.002, 0.002). The expression of hPTTG1 was significantly associated with clinical stages and lymph node metastasis status ( P =0.025, 0.007) but not with the histological grade ( P =0.144). The expression of c Myc was significantly correlated with lymph node metastasis status( P =0.032), but not with histological grades and clinical stages ( P =0.127, 0.198). There was a significant relation between the expression of hPTTG1 and c Myc ( r= 0. 463, P = 0.002). Neither of the expressions had relation with age, sex, tumor type and cholelithiasis. CONCLUSION: hPTTG1 and c Myc may play a pivotal role in the tumorigenesis and tumor progression in gallbladder carcinoma. They can be used as important markers of invasiveness of gallbladder carcinoma and may provide new targets for therapy of gallbladder cancer.
