氡及其子体对大鼠靶器官损伤的研究

摘    要:目的 研究不同剂量氡暴露致大鼠肺和外周血损伤的生物学效应,探讨二者之间的相关性。方法 雄性SD大鼠吸入氡及子体的累积剂量分别达66、111、174工作水平月(WLM)后,观察支气肺灌洗液(BALF)和外周血中细胞计数和分类的变化。测定BALF上清液和血清中LDH、GSH和总蛋白含量;用SCGE和RT-PCR方法检测BALF细胞和外周血单个核细胞(PBMC)的DNA链断裂情况及IL-6 mRNA的表达情况。结果 大鼠吸入氧及子体后,各染毒组BALF中淋巴细胞比例与对照组相比明显减少,粒细胞增多,而外周血中并没有发生相应的变化。最高剂量组BALF上清液总蛋白含量与对照组相比明显增高,而血清总蛋白则明显降低。各剂量组PBMC、BALF细胞的DNA迁移长度显著增加,有剂量.效应关系。BALF细胞中IL-6 mRNA的相对表达量在174WLM剂量组显著高于对照组,而PBMC在66和174WLM2个剂量组均显著高于对照组。结论 在该实验的染毒剂量下,大鼠吸入氡及子体后PBMC和BALF细胞DNA损伤之间存在正相关性,血清和BALF上清液中总蛋白含量之间存在负相关性。

Abstract:Objective To investigate biological effect of lung and blood in rats inhaled radon and its progeny,and to explore the correlation in pathological changes between the lung and blood.Methods Sprague-Dawley rats were exposed to radon and its progeny with the cumulative dose up to 66,111 or 174 WLM,respectively.LDH,GSH and total protein level were detected by kit. SCGE and RT-PCR were applied to determine DNA damage and IL-6 mRNA expression in BALF cells and PBMC.Results The proportion of lymphocyte was significantly lower and the proportion of granulocyte was higher than that of the control group.The total protein level of the 174 WLM group increased in BALF suspension,but decreased in serum.The distance of DNA migration in BALF and PBMC increased with a dose-effect relationship. A high expression level of IL-6 mRNA of BALF cells was observed in the 174 WLM,and in the PBMC this effect was observed in the 66 WLM and 174 WLM.Conclusion A positive correlation of DNA migration between BALF cells and PBMC could be derived,and a negative correlation between the total protein in BALF suspension and blood serum was observed.
