摘    要:目的 应用基因微矩阵芯片筛选胰腺癌的相关基因.方法 按一步法提取胰腺正常组织及胰腺癌组织总RNA,以包含4096个DNA的基因微矩阵芯片对正常胰腺组织及胰腺癌基因表达谱进行分析.结果 胰腺癌与正常胰腺组织中表达差别有显著意义的基因共有949条,其中胰腺癌下调基因468条,上调基因381条,按其功能分类共有15类.结论 基因微矩阵芯片可有效筛选出胰腺癌的相关基因;胰腺癌的发生发展由多个类型的基因参与调控,而非单一因素所致.

Abstract:Objective To screen the pancreatic cancer-related gene by cDNA microarray. Methods Total RNA was isolated by one-step protocol from pancreatic cancer and normal pancreatic tissue, and purified to mRNA by Oligotex. The genes in pancreatic cancer and normal pancreatic tissue were analyzed by cDNA microarray with 4 096 human genes. Results There were 849 differentially expressed genes in pancreatic cancer and normal pancreatic tissue, of which 468 were down-regulated and 381 were up-regulated in pancreatic cancer. According to function, they were divided to 15 kinds. Conclusion cDNA microarray can be used effectively to screen the pancreatic cancer-related genes, which are not regulated by any single gene. Many different kinds of genes are involved in the initiation and development of pancreatic cancer.
