益智挥发油温脾止泻作用及其机制的初步研究

目的:探索益智挥发油温脾止泻的功效及其作用机制。方法:利用番泻叶致小鼠急性腹泻,研究中药益智挥发油对小鼠腹泻潜伏期、稀便个数及稀便比重的影响;利用中药大黄致小鼠脾虚泻下模型,对益智挥发油改善脾虚状态和影响胃肠运动进行研究;检测小鼠血浆中脾虚状态相关的酶的水平和胃肠运动相关的NO水平和胃肠激素GAS、MOT和SS,对益智挥发油的作用机制进行研究。结果:益智挥发油0.278、0.833mL/kg剂量组能够显著的延长小鼠的腹泻潜伏期,与模型组相比,差异具有统计学意义(P<0.01);益智挥发油在一定程度上改善脾虚状态下的小鼠体重、体温以及摄食量,对炭末推进百分率无影响,但是益智挥发油0.278、0.833mL/kg剂量组能够显著的提高小鼠胃残留率,升高血浆LDH水平,降低血浆MOT和SS水平。结论:益智挥发油可以通过升高血浆LDH水平改善脾虚状态,同时降低血浆MOT和SS激素水平抑制胃肠运动达到温脾止泻的功效。

Preliminary study on warm spleen and antidiarrheal activity of volatile oil extracted from Alpinia oxyphylla

Objective: To conduct a preliminary study on antidiarrheal activity of volatile oil extracted from Alpinia oxyphylla.Methods: Mouse model of acute diarrhea induced by senna-leaf was used to study the warm spleen and anti-diarrhea effect of volatile oil from Alpinia oxyphylla.Detection indicators included the onset time of diarrhea,wet faeces number and wet faeces proportion were recorded.The mouse model of diarrhea of splenic deficiency induced by rhubarb was used to asses the effect of volatile oil on the spleen asthenia syndrome and gastrointestinal motility,detecting plasma asthenia related enzyme and gastrointestinal motility related NO level and gastrointestinal hormone GAS,MOT and SS to explore the potential mechanism.Results: Volatile oil at the dosage of 0.278 mL/kg and 0.833 mL/kg could significantly prolong the onset time of diarrhea(P<0.01) compared with the model;Volatile oil could improve the body weight,temperature and food consumption of mouse with spleen asthenia syndrome;volatile oil at dosage of 0.278 mL/kg and 0.833 mL/kg also increased the gastric residual rate,increase LDH and reduce the level of MOT and SS.Conclusions: These results suggested that volatile oil extracted from Alpinia oxyphylla might exhibit its wenpi anti-diarrhea effects partially by increasing LDH and reducing the level of gastrointestinal hormone MOT and SS.
