Abstract:Objective To study the effect of homocysteine on SIRT1 expression in vascular endothelial cells,and explore the effect of up-regulation of SIRT1 on Hcy-induced apoptosis in vascular endothelial cells.Methods The cell activity was detected by CCK8 kit,the apoptosis was detected by flow cytometry,the mRNA expression was detected by qPCR,and the protein expression was detected by Western blot.Results Homocysteine induced apoptosis of vascular endothelial cells and inhibited SIRT1 expression in a dose-dependent manner.Up regulation of SIRT1 expression in vascular endothelial cells not only significantly reduced homocysteine induced apoptosis of vascular endothelial cells(P<0.001),but also significantly inhibited the expression of Pro apoptotic gene BCL2(P<0.001)and significantly promoted the expression of anti apoptotic genes Bax and Cyt C(P<0.001).Conclusion Homocysteine inhibits the expression of SIRT1 in vascular endothelial cells,and up-regulation of SIRT1 expression can reduce homocysteine-induced apoptosis of vascular endothelial cells.
