Association between endothelial cell-and platelet-derived microparticles and outcomes of radiofrequency ablation in patients with nonvalvular AF

Abstract:Objective To analyze the effects of platelet activation and endothelial dysfunction in the posterior circulation of patients with nonvalvular atrial fibrillation(NVAF)undergoing radiofrequency ablation(RFA).Methods Venous blood samples were drawn from 35 NVAF patients(AF-Pre group and AF-Post group)and 30 normal sinus rhythm(NSR control group)controls from October 2018 to October 2019.Activation,mitochondrialΔψm depolarization of platelets and circulating platelet and endothelial-derived MPs were measured by flow cytometry.Plasma levels of von Willebrand factor(vWF)and endothelin-1(ET-1)were observed by ELISA.The relationship between PMPs,EMPs and CHA2DS2-VASc scores of NVAF patients was assessed.Results Obvious activation and increased mitochondrialΔψm depolarization of platelets were found in AF-Pre group,compared to NSR controls(P=0.0004,P<0.0001).AF-Post group had significant lower platelet activation and mitochondrialΔψm depolarization than that in AF-Pre group(P=0.0126,P<0.0001).PMPs,EMPs,vWF and ET significantly increased in AF-Pre group when compared to AF-Post group or NSR controls(P<0.0001,respectively).Quantities of PMPs(r=0.6725,P<0.0001)but not EMPs(r=0.1216,P=0.4864)were positively correlated with CHA2DS2-VASc score.Conclusions NVAF accompanied with platelet abnormal activation and apoptosis and endothelial dysfunction.Increased PMPs released from activated platelets may predict the thrombus in prognosis of NVAF.
