Functional status of T cells in children with Henoch-Schonlein purpura

Objective To observe the states of balance between TH1 / TH2 andthe function of T cells in children with Hench-Schnlein purpura (HSP) in the acute phase, through investigating the TH1 type and TH2 type cytokines levels and membrane CD30 expression on PBMC, in order to explore the pathogenesis. Methods The CD30 expressing cell counting in PBMC was performed by flow cytometry. IgA, IgG, IgM, IgE， subsets (G1～4) and cytokines were determined by radial immunodiffusion and ELISA. Results The results of the determinations are as follows:serum IgA （1.6±0.5）,controls （1.0±0.5 ）g/L] and IgE (4.4±3.5)，controls（1.1±0.7）mg/L] levels, and IgG subsets（Mean）G1 1.20 ， controls 0.15 mg/L；G2 1.13，controls 0.12，mg/L；G3 0.34，controls 0.02 mg/L；G4 0.17，controls 0.014 mg/L）；IgE 0.05,controls 0.03 mg/L）； IL-4 33，controls 16 ng/L；IL-5 (47，controls 10 ng/L）and IL-6 498，controls 310 ng/L，levels in the supernatants of cultured PBMC and CD30 expression on the PBMC of HSP patients were increased significantly in contrast to the normal controls. IL-2 (137，controls 1 537 ng/L） and IFN-γ (395，controls 721 ng/L) of HSP patients were lower. The raised IL-5 and IL-4 levels in the supernatants of cultured PBMC of HSP patients positively correlated with the increased levels of IgA（r=0.63）and IgE（r=0.87） in serum.Conclusion The function of TH1 was decreased while the function of TH 2 was increased in HSP, which were related to the pathogenesis of the disease.
