Abstract:Objective To investigate the therapeutic effectiveness of Codonopsis Pilosula Polysaccharide on mice with anemia induced by Cyclophosphamide(CTX).Methods A total of 60 SPF healthy male BALB/c mice were equally divided into 6 groups blank group,model group,low,medium and high dose groups and positive control group.Except for blank group,the other 5 groups were intraperitoneally injected with CTX by the dose of 100 mg/kg on the first day of the experiment for once a day.The blank group was intraperitoneally injected with the equal volume of physiological saline for 7 d continuously.At 2 h after CTX injection on the second day of the experiment beginning,the low,medium and high dose groups were given 10,20 and 40 ml/kg Codonopsis Pilosula Polysaccharide by gavage respectively,while positive control group was given 400 mg/kg Shengxuening Solution by gavage once a day for 7 d continuously.Levels of red blood cells(RBC),hemoglobin(Hb)and hematocrit(HCT)were detected by automatic blood cell counter,and levels of erythropoietin(EPO)and multi colony stimulating factor(multi-CSF)were detected by enzyme linked immunosorbent assay(ELISA),and levels of CD71+and Ter119+in peripheral blood were detected by flow cytometry in each group.Results RBC,Hb and HCT levels in peripheral blood in model group were significantly lower than those in blank group(P<0.05).RBC,Hb and HCT levels in low,medium and high dose groups were significantly increased compared with those in model group(P<0.05);RBC,Hb and HCT levels in low and medium dose groups were significantly lower than those in positive control group(P<0.01).Levels of multi-CSF and EPO in peripheral blood in model group were significantly lower than those in blank group(P<0.05).Levels of multi-CSF and EPO in low,medium and high dose groups were significantly increased compared with those in model group(P<0.05).The multi-CSF levels in low,medium and high dose groups were significantly lower than that in positive control group(P<0.01),and multi-CSF level was increased with increasing intervention dose.EPO levels in low and medium dose groups were significantly lower than that in positive control group(P<0.01).Levels of CD71+and Ter119+in model group were significantly lower than those in blank group(P<0.01).Levels of CD71+and Ter119+in low,medium and high dose groups were significantly increased compared with those in model group(P<0.01).Levels of CD71+and Ter119+in low and medium dose groups were significantly lower than those in positive control group(P<0.01).Conclusion Codonopsis Pilosula Polysaccharide may effectively improve mice with anemia induced by CTX.
