Abstract:Objective To observe the effect of hypertonic saline solution on blood-brain barrier(BBB)after cerebral resuscitation in rats with cardiac arrest.Methods A total of 72 SD rats were randomly divided into normal control group(S group,n=18),model group(MS group,n=18),hypertonic saline solution group(3%Sodium Chloride Solution by intravenous injection,HS group,n=18)and intravenous injection of 0.9%Sodium Chloride Solution(NS group,n=18).MS group,NS group and HS group were treated with modified asphyxia cardiac arrest to establish acute cerebral ischemia models,and then cardiopulmonary resuscitation was performed.S group and MS group did not receive intervention.HS group was infused with hypertonic saline solution(3%Sodium Chloride Solution,4ml/kg),and NS group was infused with the same dose of 0.9%Sodium Chloride Solution,and both groups were treated for 8 min consecutively.BBB leakage conditions,brain water content(BWC),as well as protein and mRNA expressions of occludin,claudin-5 and tight junction protein-1(ZO-1)after ischemia-reperfusion for 6 h and 24 h were observed.Results BBB leakage conditions and BWC levels after cerebral ischemia-reperfusion for 6 h and 24 h in MS group,HS group and NS group were worsened or increased compared with those in S group,while the above indicators in HS group and NS group were alleviated or lower compared with those in MS group,and the above indicators in HS group were also alleviated or lower compared with those in NS group(P<0.05).The decrease conditions of BBB leakage and BWC after ischemia-reperfusion for 24 h was less or lower than those after ischemia-reperfusion for 6 h(P<0.01).Compared with those in S group,protein and mRNA expressions of occludin and ZO-1 in brain tissues were down-regulated,while protein and mRNA expressions of claudin-5 were up-regulated after cerebral ischemia-reperfusion for 6 h and 24 h in MS group,HS group and NS group(P<0.05).Compared with those in MS group,protein and mRNA expressions of occludin and ZO-1 in brain tissues were up-regulated,while protein and mRNA expressions of claudin-5 were down-regulated in HS group and NS group(P<0.05).Compared with those in NS group,protein and mRNA expressions of occludin and ZO-1 in brain tissues were up-regulated,while protein and mRNA expressions of claudin-5 were down-regulated in HS group(P<0.05);the degrees of down-regulated mRNA expressions of occludin,ZO-1 and claudin-5 in brain tissues were greater with prolonged time of reperfusion(P<0.05).Conclusion Hypertonic saline solution may significantly improve brain edema after cerebral resuscitation in rats with cardiac arrest,and its mechanism may be related to improving the function of BBB.
