来氟米特对子宫内膜异位症大鼠模型作用的实验研究

摘    要:目的:观察来氟米特对子宫内膜异位症大鼠模型病灶生长行为的影响,为子宫内膜异位症的免疫治疗提供依据。方法:采用自体移植法建立大鼠子宫内膜异位症模型。移植第3周治疗组给予来氟米特35 mg/kg.d,对照组给予生理盐水1 m l/kg.d,连用3周;测量用药前后病灶体积,并采用放免法检测TNF-α水平,免疫组化法检测NF-κB的表达情况。结果:治疗组病灶体积显著低于对照组(P<0.05),组织病理学显示治疗组的异位内膜生长明显受抑制或变性坏死,病灶组织血供不足;治疗组TNF-α及NF-κB表达显著低于对照组(P<0.05)。结论:来氟米特对大鼠子宫内膜异位病灶的生长有明显抑制作用,证实免疫调节治疗子宫内膜异位症的策略具有可行性。

Abstract:Objective:To observe the effect of leflunomide on focal growth behaviors of rat models of endometriosis,provide a basis for immune therapy of endometriosis.Methods:Autologous transplantation was used to established rat models of endometriosis;on the third week after transplantation,the rats in treatment group were treated with leflunomide,35 mg·kg-1·d-1 for three weeks;the rats in control group were treated with normal saline,1 ml·kg-1·d-1 for three weeks;the focal volumes before and after treatment were measured,radioimmunoassay was used to detect the expression of TNF-α,and immunohistochemical method was used to detect the expression of NF-κB.Results:The focal volume of treatment group was significantly smaller than that of control group(P<0.05);histopathological results showed that the growth of ectopic endometrium in treatment group was inhibited or necrosis,the blood supply of focal tissues was inadequate;the expression levels of TNF-α and NF-κB in treatment group were significantly lower than those in control group(P<0.05).Conclusion:Leflunomide can inhibit the focal growth of endometrium in rat models,it is feasible to treat endometriosis by immune regulation.
