阴道毛滴虫十堰分离株对灭滴灵敏感性的实验研究

摘    要:目的:实验观察阴道毛滴虫十堰分离株对灭滴灵的敏感性。方法:收集阴道毛滴虫临床分离株,体外传代培养4代,培养11管至对数生长期,调整虫体浓度为0.5×106个/ml,计算加入甲硝唑,使终浓度分别为:62.50μg/ml、50.00μg/ml、31.25μg/ml、25.00μg/ml、15.63μg/ml、7.81μg/ml、3.91μg/ml、1.95μg/ml、0.98μg/ml、0.49μg/ml、0μg/ml,置37℃培养,分别于2、4、6、8、12、24 h观察虫体生长存活情况,计算高倍镜下虫体数目和死亡率。结果:62.50μg/ml和50.00μg/ml浓度组,滴虫死亡率均为100.00%;作用后2 h,15.63μg/ml浓度组死亡率为7.69%,31.25μg/ml组死亡率为65.51%;作用后4 h,7.81μg/ml浓度组死亡率为78.94%,3.91μg/ml浓度组死亡率为18.75%;作用后6 h,3.91μg/ml浓度组死亡率为73.68%,1.95μg/ml浓度组死亡率为21.52%;24 h滴虫最低致死量为3.91μg/ml。结论:阴道毛滴虫十堰分离株对灭滴灵较敏感,体外有效最低杀虫浓度为3.91μg/ml。

Abstract:Objective:To observe the sensitivity of isolated strains of trichomonas vaginalis in Shiyan city to metronidazole.Methods:The clinical isolated strains of trichomonas vaginalis were collected,cultured for four generations in vitro,the clinical isolated strains in 11 tubes were cultivated to exponential phase,then the concentration of trichomonas vaginalis was regulated at 0.5 ×106/ml,metronidazole was added into the tubes,the terminal concentrations were 62.50 μg/ml,50.00 μg/ml,31.25 μg/ml,25.00 μg/ml,15.63 μg/ml,7.81 μg/ml,3.91 μg/ml,1.95 μg/ml,0.98 μg/ml,0.49 μg/ml and 0 μg/ml,the clinical isolated strains of trichomonas vaginalis were cultivated at 37℃,then the survival conditions of trichomonas vaginalis were observed at 2,4,6,8,12 and 24 hours;the amount and death rates of trichomonas vaginalis were calculated under high power lens.Results:62.50 μg/ml group and 50.00 μg/ml group: the death rates of trichomonas vaginalis were 100.00%;after treatment for 2 hours,the death rates in 15.63 μg/ml group and 31.25 μg/ml group were 7.69% and 65.51%,respectively;after treatment for 4 hours,the death rates in 7.81 μg/ml group and 3.91 μg/ml group were 78.94% and 18.75%,respectively;after treatment for 6 hours,the death rates in 3.91 μg/ml group and 1.95μg/ml group were 73.68% and 21.52%,respectively;the minimum lethal concentration(MLC) of trichomonas vaginalis at 24 hours was 3.91 μg/ml.
