银杏叶总黄酮对急性肾缺血再灌注损伤大鼠的肾保护作用及机制

摘    要:目的探讨银杏叶总黄酮(TFG)对急性肾缺血再灌注损伤(IRI)大鼠的肾保护作用及可能机制。方法将30只SD大鼠随机分为假手术组、模型组及TFG低、中、高剂量组,每组6只。TFG低、中、高剂量组分别予TFG50、100和200mg/kg灌胃,每日1次,连续3d。末次给药12h后模型组及TFG各组采用夹闭双侧肾动脉45min、再灌注24h制备急性肾IRI模型。采用化学方法测定各组血清SCr和BUN水平;镜下观察肾组织形态和细胞超微结构变化;采用paller法进行肾小管损伤评分;原位末端标记法检测细胞凋亡指数(AI)。结果模型组SCr和BUN水平、肾小管损伤评分及AI均明显高于假手术组(P〈0.01);TFG各组上述指标明显低于模型组(P〈0.05或0.01),肾组织病理性损伤明显减轻。结论 TFG能明显减轻肾IRI;其机制可能与其抑制肾小管上皮细胞凋亡有关。

Abstract:Objective To investigate the effect of total flavone of gingko( TFG) against renal ischemia reperfusion injury ( IRI) in rats. Methods Totally 30 rats were randomly divided into five groups,with 6 rats in each: sham group, model group and TFG low,middle,high dose groups. TFG low,middle,high dose groups were given 50,100 and 200 mg/kg by intragastric administration ,once everyday for 3 days. Twelve hours after the last administration ,the IRI models were established in the model group and TFG groups through clamping bilateral renal arteries for 45 mins and then reperfusion for 24 hs. Serum creatinine ( SCr) and blood urea nitrogen ( BUN) were detected by chemical method; pathologic and ultrastructural changes were examined by optic and electron microscopy; the scores of renal tubule injure were estimated by paller method; renal cell apoptotic index ( AI) was examined by means of terminal deoxynucleotidyl transferase-mediated d-UTP nick end labeling. Results Compared with the sham group,the levels of SCr and BUN,the cell AI and the scores of renal tubule injure of the model group increased significantly ( P 0. 01) ; the aboved indexes of the TFG groups were significantly Lower than those of the model group ( P 0. 05 or 0. 01) ,and pathologic injure lightened. Conclusions TFG has significantly protective effect on renal IRI ,the mechanism maybe associated with suppressing apoptosis of tubular epithelial cells.
