不同方式诱导的大鼠骨质疏松模型的骨密度及骨代谢指标的差异

摘    要:目的:探讨维甲酸、D-半乳糖、苯妥英钠和糖皮质激素4种方式诱导大鼠骨质增生模型的骨密度和各项骨代谢生化指标的变化。方法:将50只大鼠随机分为对照组、维甲酸诱导模型组、D-半乳糖诱导模型组、苯妥英钠模型组和糖皮质激素模型组,每组10只大鼠。采用相应的方式诱导大鼠骨质增生,持续给药2个月后观察1个月。检测5组大鼠的骨密度、血钙、血磷、骨钙素(BGP)、碱性磷酸酶(ALP)、抗酒石酸酸性磷酸酶(TRAP)、甲状旁腺激素(PTH)、胰岛素生长因子-1(IGF-1)等。结果:股骨骨密度、胫骨骨密度及BGP、IGF-1维甲酸诱导模型组、D-半乳糖诱导模型组、苯妥英钠模型组、糖皮质激素模型组均低于对照组(P0.05)。血钙及血磷D-半乳糖诱导模型组、苯妥英钠模型组、糖皮质激素模型组低于对照组(P0.05);维甲酸诱导模型组高于对照组(P0.05)。ALP甲酸诱导模型组高于对照组(P0.05)、BGP、IGF-1维甲酸诱导模型组、D-半乳糖诱导模型组、苯妥英钠模型组、糖皮质激素模型组低于对照组(P0.05)。PTH、TRAP维甲酸诱导模型组、D-半乳糖诱导模型组、苯妥英钠模型组、糖皮质激素模型组均高于对照组(P0.05)。结论:维甲酸、D-半乳糖、苯妥英钠和糖皮质激素4种方式诱导的大鼠骨质疏松模型中,骨密度、BGP、IGF-1、PTH和TRAP表现出了相同的变化趋势,而血钙、血磷、ALP的变化趋势则因诱导方式不同而不同。

Differences in Bone Mineral Density and Bone Metabolism of Rats Model with Osteoporosis Induced by Different Methods
Abstract:Objective:To explore changes of the bone mineral density (BMD) and bone metabolism of rats model with osteoporosis induced by retinoic acid,D-galactose,phenytoin and glucocorticoids.Methods:All 50 rats were randomly divided into the control group,retinoic acid induced model group,D-galactose induced model group,phenytoin induced model group and glucocorticoids induced model group,each group ten rats.Rats model with osteoporosis was induced by the corresponding methods,given continuous administration two months and observed for one month.BMD,blood calcium,phosphorus,BGP,ALP,TRAP,PTH and IGF-1 of the rats in five groups were detected.Results:Femoral BMD,tibial BMD,BGP and IGF-1 in retinoic acid induced model group,D-galactose induced model group,phenytoin induced model group and glucocorticoids induced model group were lower than these of the control group(P<0.05).Blood calcium and phosphorus in D-galactose induced model group,phenytoin induced model group and glucocorticoids induced model group were lower than these of the control group(P<0.05);retinoic acid induced model group were higher than the control group (P<0.05).ALP formic acid induced model group were higher than the control group (P<0.05),retinoic acid induced model group,D-galactose induced model group,phenytoin induced model group and glucocorticoids induced model group were lower than the control group in BGP and IGF-1 (P<0.05).PTH and TRAP of retinotic acid induced model group,D-galactose induced model group,phenytoin induced model group and glucocorticoids induced model group were higher than these of the control group(P<0.05).Conclusion:In rats model with osteoporosis induced by retinoic acid,D-galactose,phenytoin and glucocorticoids,BMD,BGP,IGF-1,PTH and TRAP manifest the same change trend,while the change trends of blood calcium,phosphorus and ALP are different because of different induction methods.
